Determination of simple heritabilities using maize (Zea mays) as an example, basics of Drosophila genetics; chromosome number, balancers; the P element and other transposons in Drosophila; specific application of the P element: the GAL4/UAS system; further modifications: Recombination using FRT/FLP; targeted generation of deficiencies for individual genes (phi/Cre/Lox).
Independent performance of experiments and acquisition of knowledge on Drosophila strain maintenance, male/female differentiation, fly handling; identification and recognition of balancer strains based on phenotypic markers; set-up and analyses of crossing plans; Determination of chromosomal localization of transgenes; Crossing with balancer strains; Jump-out experiments to produce mutants of single genes; FRT/FLP crosses to generate genomic deficiencies; Meiotic recombination to generate double mutants, resp. Labeling of chromosomes using multimarker techniques; GAL4-UAS crosses for analysis of mutant phenotypes, evaluation of adult structures (eye, wing, etc.); analysis of mutants using in situ hybridization/antibody staining.
Qualification Goals
Students should acquire basic knowledge of Drosophila genetics that goes well beyond the knowledge of classical genetics. In particular, the targeted use of transposons for the analysis and generation of specific mutants will be taught in this module. Here, in addition to the theoretical basics, independent practical skills in the design and execution of crossing experiments will be taught in particular, as well as the detailed presentation of a crossing plan and the critical evaluation of the data.
